detection, virus, rna. - The polymerase chain reaction (PCR) is increasingly being used for the rapid detection of viral pathogens in medical samples. It is an in vitro-technique for amplification of specific genomic fragments from desoxyribonucleic acid (DNA). For detection of genome fragments from viruses with RNA genomes, the complementary DNA (cDNA) has to be synthesized by reverse transcription (RT-PCR). A prerequisite for the performance of RT-PCR is the accessibility of the RNA for the corresponding enzymes and reagents. In addition, to avoid false negative results, the RNA has to be protected from degradation and other damage, and substances that inhibit the enzymatic reactions have to be removed.
