Farbdopplersonographische Untersuchungen zur Funktion des bovinen Corpus luteum

The objective of this thesis was to determine if cyclic changes of luteal blood flow (LBF) are correlated with those of luteal size (LS) and plasma progesterone concentration (P4). Furthermore, it was examined if during the mid of diestrus individual differences in these parameters are related with each other. To examine cyclic changes, corpora lutea of 10 synchronized lactating Holstein Friesian cows were estimated by transrectal sonography on days 4, 5, 6, 7, 8, 10, 12, 14, 16, -5, -4, -3, -2, and -1 (Day 1 = ovulation). Additionally, after each investigation blood samples were collected to analyze P4 by using EIA. Luteal size was quantified by measuring the cross-sectional area on B-mode images of the Corpus luteum (CL) and LBF was determined by calculating the maximum area of colour pixels on digitalized images of the CL. Luteal size doubled (P ≤ 0.05) during the luteal growth phase (until Day 7), remained at a high level during the luteal static phase (Day 8 to 14), before decreasing progressively by nearly 53 % until Day -1. Changes of LBF reflected P4 concentrations during the estrous cycle more reliable than LS, i. e. LBF increased during the growth phase by approx. 133 %, doubled during the static phase and decreased rapidly during luteal regression by nearly 90 % like P4. Values of luteal size > 4.5 cm² (sensitivity: 42 %; specificity: 100 %; +PV: 100%; -PV: 33%) and LBF values > 0.6 cm² (sensitivity: 69 %; specificity: 100 %; +PV: 100%; -PV: 48%) were indicative for P4 levels > 1 ng/ml. The aim of the second study was to analyse if there are differences in peripheral P4 concentration, LS and LBF between cows and if there are, if these are related with each other during the mid of diestrus. For this study, examinations were carried out in 31 lactating Holstein Friesian cows on Day 10, 11, 12 and 13. The determined sonographic and endocrinically parameters were the same as in the first study. For the investigation if there are relationships between P4, LS and LBF for each cow mean values of all investigated Days were calculated. All three parameters exhibited strong individual differences (min-max CV: 19.0 - 26.5%). P4 and luteal size (r = 0.25; P = 0.18) and P4 and luteal blood flow (r = 0.26; P = 0.15) were not correlated, but there was a moderate correlation between blood flow and luteal size (r = 0.41; P = 0.02).   The results of the two studies show that luteal blood flow is more reliable for the evaluation of cyclic changes of the luteal function in cows than luteal size, but no one of both parameters allows conclusions to the absolute peripheral progesterone level during the mid of diestrus.

Ziel dieser Dissertation war es zu untersuchen, ob zyklusbedingte Veränderungen im lutealen Blutfluss (LBF) mit denen der lutealen Größe (LS) und der Plasmaprogesteronkonzentration (P4) korrelieren und ob Zusammenhänge zwischen den individuellen Unterschieden in diesen Parametern während des mittleren Diöstrus bestehen. Um die zyklusbedingte Veränderungen zu verfolgen, wurden die Corpora lutea von 10 Holstein-Friesian Kühen an den Zyklustagen 4, 5, 6, 7, 8, 10, 12, 14, 16, -5, -4, -3, -2 und -1 (Tag 1 = Ovulation) sonographisch untersucht. Zusätzlich wurde im Anschluss an die jeweiligen Untersuchungen eine Blutprobe zur Bestimmung von P4 mittels EIA entnommen. Die LS wurde anhand der lutealen Querschnittsfläche im B-Mode und LBF anhand der farbigen Fläche des im Dopplermodus aufgenommenen Corpus luteum Querschnittes ermittelt. Die Größe des Corpus luteum verdoppelte (P ≤ 0.05) sich während der Wachstumsphase (Tag 4 bis 7), blieb während der mittleren Phase des Diöstrus auf konstant hohem Niveau (Tag 8 bis 14), um anschließend in der Regressionsphase (Tag -5 bis -1) kontinuierlich um ungefähr 53 % abzufallen. Die Veränderungen im LBF spiegelten diejenigen der P4-Konzentration während des Zyklus wieder. Das heißt, die luteale Durchblutung stieg während der Wachstumsphase um ca. 133 % an, verdoppelte sich während der mittleren Phase des Diöstrus und fiel während der Regressionsphase um ungefähr 90 % ab. LS-Werte > 4.5 cm² (Sensitivität: 42%; Spezifität: 100%; +PW: 100%; -PW: 33%) und LBF-Werte von > 0.6 cm² (Sensitivität: 69%; Spezifität: 100 %; +PW: 100%; -PW: 48%) wiesen auf P4-Level > 1 ng/ml hin. Im Rahmen der zweiten Studie wurde überprüft, ob es individuelle Variabilitäten in den LS- und LBF-Werten gibt und falls ja, inwieweit diese mit Unterschieden in der P4-Konzentration zwischen den Tieren zusammenhängen. Dazu wurden 31 Holstein-Friesian Kühe an den Zyklustagen 10, 11, 12 und 13 untersucht. Es wurden dieselben sonographischen und endokrinologischen Parameter wie im ersten Versuch ausgewertet. Um etwaige Zusammenhänge zwischen P4, LS und LBF zu bestimmen, wurde für jede Kuh der Mittelwert der Parameter aus allen Untersu-chungstagen herangezogen. Alle drei Parameter wiesen starke individuelle Unterschiede auf (min-max CV: 19.0-26.5%). Der luteale Blutfluss korrelierte moderat mit LS (r = 0.41; P = 0.02), aber weder LS  (r = 0.25; P = 0.18) noch LBF (r = 0.26; P = 0.15) hingen mit P4 zusammen. Die Ergebnisse der beiden Studien zeigen, dass der luteale Blutfluss besser geeignet ist als die Größe, um zyklusbedingte Veränderungen in der Funktion des Corpus luteum nachzuweisen. Keiner der beiden sonographisch ermittelten Parameter erlaubt jedoch eine Aussage über den absoluten peripheren Progesterongehalt in der Mitte des Diöstrus beim Rind.
